Une présence humaine à Monthou-sur-cher est certaine depuis au moins le Ier siècle puisqu'au au lieu-dit le Moulin du Ru, en rive droite du Cher et dans la partie sud de la commune, en limite de Thésée, se trouve une partie de l'agglomération antique de Tasciaca. Le patrimoine architectural de la commune comprend deux bâtiments protégés au titre des monuments historiques, le château du Gué-Péan, classé en 1980, et l'église Saint-Cyr de Monthou-sur-Cher, inscrite en 1926.
